RP-HPLC法测定散瘀软膏中盐酸小檗碱的含量

摘　　要：[目的]建立散瘀软膏中盐酸小檗碱的HPLC含量测定方法。[方法]采用反相离子对高效液相色谱法。色谱柱:Agilent Zorbax Eclipse XDB C8(4.6×150mm,5μm);流动相:乙腈-0.1%磷酸溶液=50∶50(每100mL流动相加入十二烷基硫酸钠0.1g);检测波长:265nm;流速:1.0mL·min-1;室温:20℃;进样量:20μL。[结果]盐酸小檗碱在39.40～394.0ng的范围内线性关系良好,回归方程为:Y=4.073×103x+2.248×103(r=0.9992);加样回收率在97.9～100.9%,平均回收率为99.4%,RSD为1.5%。[结论]所采用的定量方法简便、可靠,结果准确,重复性良好,可用于散瘀软膏的质量控制。[Objective] To establish a content determination method of Berberine hydrochloride in San Yu Ointment by HPLC.[Methods] RP-HPLC was performed to determine Berberine hydrochloride on a column of Agilent Zorbax Eclipse XDB-C8(4.6×150mm,5μm)at 20℃; Acetonitrile-0.1 % H3PO4 (50∶50)(Adding 0.1 g C12H25NaO4S to every 100 mL)was used as mobile phase,the flow rate was 1.0 mL·min-1.The detection wavelength was at 265 nm.[Results]A good linearity can be found in the range of 39.40 ~394.0 ng,and the regression equation was Y = 4.073×103x+2.248 × 103 (r=0.9992).The recovery was between 97.9 %~100.9 %,and the average recovery rate was 99.4 %,RSD was 1.5 %.[Conclusion]The method is simple,rapid,accurate and available,which can be used to control the quality of San Yu Ointment.
