摘 要:选用HYPERSILC18柱 ,以乙腈 水 ((5 0∶5 0 ) (0~ 4 8min)→ (6 5∶35 ) (48~ 6 8min)→ (75∶2 5 ) (6 8~ 95min) ) (0 .1%乙酸 )为流动相进行高效液相分离 ,二极管阵列 (DAD)和电喷雾质谱 (ESI MS)两种检测方法在线检测。经过分析紫外图谱和质谱 ,并与文献进行比较 ,鉴定了 15个茯苓三萜化学成分 ,并推测了其余的8个三萜化学成分。应用HPLC/DAD/ESI MS方法可快速有效地鉴定茯苓中的三萜化学成分。A method for the identification of triterpene constituents of Poria cocos (Schw.) Wolf. was developed by high performance liquid chromatography/diode array detection/mass spectrometry (HPLC/DAD/ESI-MS). A HYPERSIL C-18 column was applied in the HPLC analysis of the extract of Poria cocos with acetortitrile-water ( (50:50) (0 similar to 48 min) -->(65:35) (48 similar to 68 min) --> (75:25) (68 similar to 95 min)) (0.1 % acetic acid) as mobile phase. The elutes was detected by diode array detection (DAD) to give the chromatogram. and ultraviolet (UV) spectra of the peaks in the chromatogram. And on-line electrospray ionization (ESI-MS) detection was performed to obtain the MS spectra. Then, fifteen triterpenes were identified by the analysis of the UV and ESI-MS spectra as well as the comparison with that reported in literatures, and eight structures of other triterpenes were deduced. Consequently, HPLC/DAD/ESI-MS can be performed to identify the triterpene constituents of Poria cocos quickly and effectively.
