摘  要:将叶绿体处理后用聚乙烯醇 苯乙烯吡啶 (PVA SbQ)聚合膜进行固定 ,得到固定化叶绿体膜 ,该膜中叶绿体束缚酶能催化H2 O2 的分解产生氧 ,而除草剂对该指示反应有抑制作用。利用氧电极 ,建立了新的用于检测痕量除草剂的电流分析法。所检测的除草剂有阿特拉津、莠灭净、草净津、西玛三嗪、扑草净、敌草隆和百草枯等 ,检测浓度范围分别为 0 .2 5~ 1.75 ,0 .0 4~ 0 .7,0 .1~ 1.1,0 .1~ 0 .8,0 .2~ 1.8,0 .0 1~ 0 .12和0 0 0 6~ 0 .0 2mg/L。研究了PVA SbQ光聚合物固定化叶绿体膜的性质 ,结果证明该法固定的叶绿体束缚酶具有活性高、寿命长和使用方便等特点。该方法用于测定样品中除草剂残留物 ,取得了满意的结果。文中还对叶绿体中活性酶的作用机理进行了探讨。A novel amperometric method for the determination of herbicide was proposed. It is based on the inhibition of herbicides on the decomposition of hydrogen peroxide catalyzed by the chloroplast-bound enzymes, then the oxygen was produced in the cell and detected using a clark electrode. The chloroplast isolated from spinach leaves was immobilized by entrapment in a membrane of polyvinylalcohol bearing styrylpyridinium groups (PVA-SbQ). Herbicides including atrazine, paraquat, diuron, ametryne, prometryne, simazine and cyanazine could be determined in concentration ranges of 0.25 similar to 1.75, 0.06 similar to 0.02, 0.01 similar to 0.12, 0.04 similar to 0.7, 0.2 similar to 1.8, 0.1 similar to 0.8 and 0.1 similar to 1.1 mg/L. The method for immobilizing chloroplast in PVA-SbQ membrane was found to be promise to make enzymes stable for several months at -20degreesC. The key enzymes in chloroplast for scavenging hydrogen peroxide were discussed. The method is sensitive, simple and convenient. Reproducible results were obtained in determination of different herbicides'. The mechanism of chloroplast-bound enzymes in the determination was discussed.
