红提葡萄组织培养的研究

摘　　要：以葡萄的茎尖为外植体,研究了不同的灭菌剂对葡萄外植体萌发的影响,并筛选出最佳消毒剂,同时筛选出不同培养基和激素浓度配比。结果表明:不同的灭菌剂对无菌苗的萌发与生长有显著的影响,使用70%乙醇灭菌30 s,0.1%升汞灭菌10 min,可以达到最佳灭菌效果。筛选出1/2MS+BA 0.5 mg/L+NAA 0.2 mg/L+蔗糖30 g/L+琼脂7 g/L,pH 5.8的培养基为外植体萌发的最适培养基配比。Taking mature embryo of grape as explants,the influence on the explants growth under different disinfectants were studied.Different mediums and hormone concentration ratios were selected.The results showed that the different disinfectants had a significant impact on the germination and gwowth of non-valline.stericizing for 30 seconds in 70% alcohol and stericizing for 10 minutes in 0.1% mercuric could achieve the best stericizing effect.The 1/2MS+BA 0.5 mg/L+NAA 0.2 mg/L+sucrose 30 g/L+agar 7 g/L medinm under pH 5.8 was the best medium for germination of matuer embryos.
