摘 要:运用RAPD分子标记技术,对12个油松天然居群中192个单株进行遗传多样性分析。用筛选出的20条引物共扩增出289个条带,其中多态性条带220个,多态性条带百分率为76.12%。用NTSYS-PC软件计算遗传相似性系数进行聚类分析。结果表明:192份油松材料间的SC值变化范围为0.5986~0.9654,平均为0.7379。采用UPGMA法,在SC值为0.76的水平上可将供试材料聚为3类。表明我国油松天然种群间产生一定程度的变异,在分子水平上呈现出遗传多态性。Random amplified polymorphic DNA(RAPD)markers were used to measure genetic diversity of 192 individuals from 12 natural populations of Pinus tabulaeformis Carr.A total of 289 loci were identified from 20 RAPD primer,among which 220 loci were polymorphic and accounted for 76.12 percent.Similar coefficients were calculated and a dendrogram was generated by unweighted pair group method of arithmetic average using NTSYS-PC.The results showed that the genetic similarity coefficients ranged from 0.598 6 to 0.965 4 with an average of 0.737 9.All the 192 materials were classified into 3 groups at level 0.76;And results revealed an abundant genetic diversity of Pinus tabulae formis Carr. natural populations in China,and most of the genetic variation existed in intrapopulation.
