摘 要:目的:研究草珊瑚不同种源间的遗传多样性及亲缘关系,并根据酶谱特征,提供种子真实性鉴定依据。方法:采用垂直板聚丙烯酰胺凝胶电泳法,对15个群落的草珊瑚种子进行酯酶同工酶测定和谱带分析,用Ntsys2.10软件计算遗传相似性系数,通过UPMA法进行聚类分析,评价不同种源的亲缘关系。结果:种子酯酶同工酶共出现12个谱带类型,分为3个区,即A区(0.17~0.36),B区(0.46~0.66)和C区(0.75~0.82)。其中Rf=0.36、Rf=0.59和Rf=0.66为3条特征酯酶谱带。草珊瑚不同种源间遗传相似度在0.50~1.00间。结论:种子酯酶同工酶谱带能大致区分的草珊瑚种子的来源,并有效地鉴定出种子真实性。草珊瑚种源亲缘关系较近,与产地没有必然联系。15批种子样品划可分为四个类群,遗传关系呈现多样性。Objective:Sarcandra glabra is a wildly-used Chinese medicinal herb and collected by Chinese Pharmacopoeia(edition of 2005).With the rapid development of the seed propagation techniques and cultivation,seed source stability is the key to guarantee the quality of the Chinese herb.To understand the germplasm resources,the genetic diversity and genetic relationship among the various provenances could be analyzed.Methods:Acrylamide Gel Electrophoresis was used to analyze the esterase isozyme of fifteen samples from seeds of Sarcandra glabra.The genetic similarity and cluster analysis were performed by Ntsys2.10 software and UPCMA method.Results:The study showed that there were twelve esterase isozyme types in fifteen samples of Sarcandra.The esterase isozyme types could be divided into three zones: A(0.17-0.36),B(0.46-0.66)和C(0.75-0.82)in which Rf=0.36、Rf=0.59 and Rf=0.66 were typical bands.Genetic similarities of fifteen provenances were ranged from 0.5-1.00.Conclusion Cluster Analysis of esterase isozyme showed that fifteen samples could be divided into 4 group.The genetic relationship of seed provenance is close,but not necessarily linked with the origin.
