摘 要:目的优选黄连解毒汤方药的提取工艺条件。方法采用半仿生法,在药材规格、煎煮温度、煎煮用水量等条件相同的情况下,用均匀设计U9(91×33)表,以盐酸小檗碱、黄芩素、栀子苷、总生物碱、总黄酮、HPLC总面积、相对分子质量≤1 000的提取物为指标,综合评价,优选该方药的提取工艺条件。结果三煎用水的pH依次为A=2.061 4,B=7.051 7,C=8.964 2;提取总时间D=4.968 0。结论结合生产实际,确定该方药用半仿生法提取的工艺条件为:3煎用水的pH值依次为2.0,7.0,9.0,提取时间依次为2.0,1.5,1.5 h。Objective To optimize technical conditions for Huanglian Jiedu decoction by semi-bionic extraction.Methods The optimum extraction conditions were comprehensively evaluated and screened by uniform design U9(91×33),maintaining the same traditional Chinese medicine specification,extraction temperature and extraction water volume.The contents of berberine,baicalin,geniposide,total alkaloid,total ?avonoid,total integral calculus area of HPLC and extract with MW ≤ 1000 Da were used as evaluating indicators.Results The optimum pH values of 3 times of decoction water were A = 2.0614,B = 7.0517 and C = 8.9642;the total extraction time was D = 4.9680.Conclusion Combined with the production practice,the optimum extraction conditions for Huanglian Jiedu decoction were as follows: pH values of 3 times of decoction water were 2.0,7.0 and 9.0;the extraction time was 2.0 h,1.5 h and 1.5 h.
