摘 要:目的探讨人低质量胚胎来源的单个卵裂球继续发育能力,并尝试建立人胚胎干细胞系。方法采用透明带溶解法将人低质量胚胎分离成单个卵裂球,再行体外培养,观察卵裂球的生长发育情况。结果 62个正常受精的人低质量胚胎经分离后共获取252个单卵裂球。卵裂球经体外培养继续卵裂率为47.6%,致密化率为28.6%,早期囊胚形成率为20.2%,晚期囊胚形成率为13.1%,滋养层细胞样生长物形成率为9.1%,无人胚胎干细胞系建立。结论人低质量胚胎的单个卵裂球通过体外培养可发育到囊胚阶段,而建立人胚胎干细胞系的方法需进一步改进。该研究提示需根据不同的培养条件来审视卵裂球全能性,为建立低质量胚胎单个卵裂球来源的人胚胎干细胞系提供了新的思路。Objective To investigate the developmental capacity of single blastomere isolated from human low-quality embryo and try to establish human embryonic stem cell lines.Methods Blastomeres were isolated from human low-quality embryos by zona pellucida digestion,the blastomeres were cultured in vitro and their development was evaluated.Results A total of 252 blastomeres were isolated from 62 normally fertilized human low-quality embryos.The cleavage rate of blastomeres was 47.6%,28.6% of the blastomeres could develop to compaction stage,20.2% of the blastomeres could develop to early blastocyst stage,13.1% of the blastomeres could develop to late blastocyst stage,9.1% of the blastomeres could develop to trophectoderm-like outgrowth and there was no derivation of human embryonic stem cell line.Conclusions The single blastomere isolated from human low-quality embryo can develop to blastocyst while human embryonic stem cell line derivation method should be modified.This investigation proves that we need to reconsider the totipotency of single blastomere according to different culture condition,and it also provides new strategies for deriving human embryonic stem cell line from single blastomere isolated from low-quality embryo in the future.
