摘 要:目的:通过测定游离脂肪酸(FFA)、白介素-6(IL-6)、干扰素(INF)等脂肪细胞因子的浓度,从而探讨西洋参活性成分对胰岛素抵抗脂肪细胞模型脂肪细胞因子分泌的影响。方法:通过高糖高胰岛素刺激,建立胰岛素抵抗脂肪细胞模型,随机分为空白组、模型组、罗格列酮组、20(R)人参皂苷Rb1组、20(R)人参皂苷Rb2组、人参皂苷R3组、拟人参皂苷F11组、西洋参复合成分组。药物干预培养24h后,检测各组脂肪细胞因子的浓度。结果:与模型组细胞FFA分泌量比较,20(R)人参皂苷Rb1组、20(R)人参皂苷Rb2组、西洋参复合成分组、罗格列酮组FFA分泌量明显降低,差异极显著(P<0.01),拟人参皂苷F11组也有显著差异(P<0.05),其余无统计学差异;与模型组细胞IL-6分泌量比较,罗格列酮组、拟人参皂苷F11组、西洋参复合成分组、20(R)人参皂苷Rb1组细胞IL-6分泌量均降低,差异极显著(P<0.01),其余无统计学差异;与模型组细胞INF分泌量比较,人参皂苷R3组、西洋参复合成分组、20(R)人参皂苷Rb1组INF分泌量明显降低,差异极显著(P<0.01),拟人参皂苷F11组、罗格列酮组也有显著差异(P<0.05),其余无统计学差异。结论:西洋参活性成分通过对胰岛素抵抗脂肪细胞模型FFA、IL-6、INF等脂肪细胞因子分泌的影响,调节脂代谢,从而达到改善胰岛素抵抗的目的。Objective:To study the effect of active constituents of American Ginseng in the treatment of insulin resistance in lipid Metabolism to survey the concentration of FFA and IL-6,INF in the cell supernatants.Methods:Induce the preadipocyte cells by high glucose and high insulin in order to modelling adipocyte insulin resistance.Then they were divided randomly into control group,model group,rogridone group,ginsenoside R3 group,20(R)ginsenoside Rb1 group,20(R)ginsenoside Rb2 group,pseudoginsenoside F11 group,compound constituents of Ginseng group.After 24 hours,survey the concentration of FFA by colorimetry and the concentration of IL-6,INF in the cell supernatants by ELISA method.Results:Compared with model group by the concentration of FFA,there was an extreme difference between 20(R)ginsenoside Rb1 group,20(R)ginsenoside Rb2 group,compound constituents of Ginseng group,roglitazone group and model group(P<0.01).There was a significant difference between pseudoginsenoside F11 group and model group(P<0.05),Others have no senses.Compared with model group by the concentration of IL-6,there was an extreme difference between pseudoginsenoside F11 group,compound constituents of Ginseng group,roglitazone group,20(R)ginsenoside Rb1 group and model group(P<0.01).Others have no senses.Compared with model group by the concentration of INF,there was an extreme difference between ginsenoside R3,20(R)ginsenoside Rb1 group,compound constituents of ginseng group and model group(P<0.01).There was a significant difference between pseudoginsenoside F11 group,roglitazone group and model group(P<0.05).Others have no senses.Conclusion:Ginsenoside R3,20(R)ginsenoside Rb1 group,20(R)ginsenoside Rb2,pseudoginsenoside F11 can affect the concentration of FFA and Cytokines in the cell supernatants which were able to improve insulin resistance.
