摘 要:目的观察高原低氧环境下骨质疏松大鼠模型的松质骨骨密度、力学性能及骨小梁结构的变化。方法选用纯种SD雌性大白鼠72只,随机分为高原去势组和平原去势组、高原对照组和平原对照组。术后分别在海拔高于3000m的高原和低于500m的平原饲养。并于术后1个月、3个月和6个月测量大鼠的股骨骨密度和腰椎生物力学强度,并行Micro-CT分析。结果 3个月时,高原和平原饲养的去势大鼠骨密度下降较为显著(P<0.05),且高原去势组骨密度明显低于平原去势组(P<0.05),对照组均无显著变化(P>0.05)。6个月时,除平原对照组骨密度无显著变化,其他三组骨密度均有显著降低,且高原去势组骨密度明显低于其他组,差异具有统计学意义(P<0.01)。生物力学检测发现,去势6个月以后,高原去势组、平原去势组、高原对照组和平原对照组抗压强度较去势后1个月分别降低29.48%、24.85%、15.63%和3.98%,高原去势组下降更为明显(P<0.01)。Mi-cro-CT分析显示,高原去势组大鼠骨的相对骨体积(BV/TV)、骨小梁数目(Tb.N)显著低于其他组,骨小梁分离度(Tb.Sp)则较其他组显著增高,差异具有统计学意义(P<0.01)。结论高原缺氧环境可引起骨代谢异常,并加速骨质疏松发展的进程,应用Micro-CT分析有助于研究高原骨质疏松动物模型的骨小梁结构变化。Objective To observe the cancellated bone density,biomechanical and microstructure of bone trabecula in ovariectomized rats with osteoporosis at high altitude.Methods 72 female SD rats were chosen and divided into group A(ovariectomized group at high altitude),group B(ovariectomized group at low altitude), group C(normal group at high altitude) and group D(normal group at low altitude).After operation,the rats were bred at high altitude and low altitude respectively.The bone density of the femoral bone,biomechanical were measured,and microstructure of bone trabecula were analyzed by micro-CT at 1,3,6 months.Results At 3 months after ovariectomy, the bone mineral density of ovariectomized rats was significantly decreased compared with the normal group(P<0.05).The bone mineral density of ovariectomized group at high altitude was lower than that of ones at low altitude(P<0.05).6 months after operation ,the bone mineral density of ovariectomized group at high altitude was significantly lower than that in other groups(P<0.01).Detection of the biomechanical:6 months after operation,the crushing strength decreased 29.48%,24.85%,15.63% and 3.98% in A,B,C,D group respectively compared with that on 3 months.The crushing strength in groups at high altitude was significantly lower as compared with those other group( P<0.01).The BV/TV and Tb.N in ovariectomized group at high altitude were significantly lower while the Tb.Sp was significantly higher (P<0.01 ). Conclusions It suggest that the effect of oxygen deficit at high altitude might cause abnormal bone metabolism and accelerate the evolution of osteoporosis. The application of the micro-CT is helpful for the study of microstructure of the cancellated bone in ovariectomized rats with osteoporosis at high altitude.
