摘 要:目的:观察小鼠胸腺树突状细胞(TDCs)的分布、含量与组织形态。方法:采用光镜、电镜和树突状细胞标志物(CD205、CD1a 与 MIDC-8)免疫组化标记方法观察小鼠胸腺切片。结果:TDC 在皮髓交界区分布最多,其次为皮质区,髓质区最少。树突状细胞约占胸腺细胞总数的2%。胸腺树突状细胞表面形成胞浆突起向周围包绕的淋巴细胞间延伸,核不规则多有凹陷,胞浆低电子密度,细胞器稀少且不含溶酶体。结论:多种树突状细胞标志物联合标记,结合光、电镜形态学观察,可以有效地记数并显示 TDCs 的典型特征。Objective:To investigative the morphology and distribution of thymus dendritic cells (TDCs)in mouse.Methods:The morphology and distribution of TDCs in mouse were observed and analyzed by the electron microscopy and the immunohistochemistry with DCs marker (including CD205,CDla and MIDC-8).Results:The results showed that TDCs were located most abundantly in the adjoining area of thymus cortex and medulla,lesser in the cortex,and least in the medulla.TDCs accounted for 2% of all cells in the thymus.On TDCs surface there were many processes which extended into diastema surrounding lymphocytes.The nucleus was irregular in shape and there were indentations.The cytoplasm was electron-lucent and there were few organelles without any lyso- some.Conclusion:Our studies suggested that with the aid of immunohistochemistry staining with various perti- nent markers as well as ultrastructure examination,it was able to dexterously count the number of TDCs and to several their morphological characters in the thymus.
