摘  要:目的:观察急性肺损伤(ALI)小鼠 T 辅助淋巴细胞(TH)TH1/TH2的平衡变化,探讨 TH1/TH2平衡变化在 ALI 发生中的作用及意义。方法:BLAB/C 小鼠随机分成 ALI 组、对照组。酶联免疫吸附法(ELISA)测定脾细胞培养上清液中干扰素-γ(IFN-γ)、白细胞介素-4(IL-4)蛋白含量,半定量逆转录聚合酶联反应(SqRT-PCR)方法进一步观察脾细胞中 IFN-γ mRNA、IL-4 mRNA 表达。伊文思蓝荧光法反映肺血管通透性改变。肺湿重/干重比反映肺水量。HE 染色观察肺病理变化。结果:与对照组比较,ALI 组脾细胞中 IFN-γ、IFN-γmRNA 表达均显著下降(P<0.01),IL-4明显升高(P<0.01),IL-4mRNA 在各组均未见表达。与对照组比较,ALI 组肺组织中伊文思蓝浸出量、肺湿重/干重比均明显升高。结论:ALI 时,TH1向 TH2漂移,提示炎症与抗炎症反应失衡,抗炎症反应增强。Objective;To investigate changes in balance of TH1/TH2 cytokines in acute lung injury and their sig- nificance.Methods;Mice were randomly divided into saline control group,and acute lung injury group (n=12 per group).By using enzyme linked immuno sorbent assay (ELISA),interferon-γ (IFN-γ) and interleukin-4 (IL-4) levels in supernatants were quantified,meanwhile cytokine messenger RNA expression was determined by semi- quantitatively reverse transcriptase polymerase chain reaction (RT-PCR).The pulmonary extravascular lung water was determined by lung wet to dry weight ratio.The quantity of protein extravasation was measured by Evan's blue fluorescence,and the change in lung pathology was examined by using HE staininging.Results;As compared with control group,splenocyte IFN-γ and IFN-γ mRNA expression were decreased (P<0.01),IL-4 was in- creased (P<0.01),while IL-4 mRNA expression was not detected under the same experimental conditions in ALI group.The lung wet to dry weight ratio and quantity of lung Evan's blue extravasation were increased markedly in ALI group.Conclusions;A shift to a TH2-type splenocyte cytokine response occurs after ALI,suggesting an im- balance of inflammatory and anti-inflammatory responses after ALI and enhancement of anti-inflammatory re- sponse.
