摘  要:目的:探讨颈动脉粥样硬化(CAS)斑块中血管内皮生长因子受体Flk-1和Flt-1表达的改变,并分析其与斑块内炎症细胞浸润和新生血管的相关性。方法:应用免疫组化和原位分子杂交方法分别检测CAS斑块中Flk-1和Flt-1蛋白和mRNA表达的改变,以CD34免疫组化染色标记斑块内新生血管。结果:Flk-1和Flt-1均主要表达于炎症细胞浸润区的巨噬细胞和单核细胞,在坏死区周边基质内,也存在不同程度的Flk-1和Fkt-1染色带,二者与炎症细胞浸润之间均具有明显的相关性(Flk-1:r=0.41,P<0.05;Flt-1:r=0.71,P<0.01);而新生血管内皮细胞仅有Flk-1表达,与新生血管之间具有明显的相关性(r=0.64,P<0.01),而无Flt-1表达。结论:在CAS病理中,血管内皮生长因子Flk-1和Flt-1均参与了炎症介导过程,而在新生血管生成中可能仅有Flk- 1的参与。Objective:To investigate changes in expression of vascular endothelial growth factor (VEGF) recep- tor Flk-1 and Flt-1 in carotid atherosclerosis (CAS) plaques,and analyze their correlations with the intraplaque in- flammatory infiltration and neoangiogenesis.Methods:Immunohistochmistry and in situ hybridization were used to detect the protein and mRNA expression of Flk-1 and Flt-1,and immunohistochmistry of CD34 was used to label the intraplaque neoangiogenesis.Results:Both Flk-1 and Flt-1 were mainly expressed in the macrophages and monocytes in the inflammatory infiltration area,and zonally distributed in the matrix surrounding the necrotic are- a.There were significant correlations between Flk-1,Flt-1 and the inflammatory cell infiltration (Flk-1:r=0.41, P<0.05;Flt-1:r=0.71,P<0.01);Only Flk-1 was expressed in the endothelium of neonatal vessels,and there was a significant correlation between Flk-1 expression and the neoangiogenesis (r=0.64,P<0.01).Conclusions: In the pathological process of the CAS,both Flk-1 and Flt-1 being VEGF receptors,might participate in the in- flammatory response,but only Flk-1 was probably involved in the neoangiogenesis.
