摘  要:目的:了解在无保护剂条件下细胞连接在组织冷冻损伤中的作用。方法:采用细胞单层中可形成细胞连接的MDCK细胞和不形成细胞连接的V79细胞,分为细胞悬液组(Suspension)、贴壁组(Single Tough)和单层细胞组(Monolayer)分别进行0-40℃之间的梯度冷冻实验,然后采用WST-1和MTT两种方法测定细胞的活力并进行比较。结果:两种方法测定的结果一致,均显示两种细胞的细胞悬液冷冻后活力高于贴壁组和单层细胞组;MDCK细胞贴壁组活力高于单层细胞组,V79细胞则无明显差异;MDCK单层细胞组活力却明显低于V79单层细胞组。结论:组织细胞间的连接以及细胞与细胞基质的连接对组织冷冻的损伤有重要的作用,它们的存在直接影响组织细胞的活力,其原因可能与构成细胞连接的细胞骨架系统有关。The main purpose of this study was to investigate the role of cell junction in the injury of tissue without any cryoprotectant after cryopreservation. Methods: Epithelial MDCK cell line , which have abili-ty to form well-defined cell junctions after eonfluency, and fibroblast V79 cell line which have not such ability, were used for graded freezing experiments, either in suspension, single tough attached to flasks or as monolayers on flasks. Then the viability was assessed by WST-1 and MTT, respectively. Result: The results of viability of two type of cells assessed by WST-1 were well-fitted with that of MTT. The recovery of two type of cells in suspen-sion was higher than that of single tough group and as monolayers. Single tough group of MDCK had higher recov-ery than its monolayers group. At the same time, such tendency was not observed on V79 cells. As compared with V79 monolayers, lower recovery was found on MDCK monolayers. Conclusion: The junction between cells and the adhension between cell and matrix have important roles on the hypothermic impairment of tissue. Their existence affect the recovery of cells under hypotherraia. The cytoskeleton system may contribute to the impairment caused by junctions.
