慢性高原病患者骨髓单个核细胞Caspase-3表达研究

摘　　要：目的:探讨慢性高原病(chronic mountain sickness,CMS)患者骨髓单个核细胞(BMMNCs)caspase-3 mRNA表达的变化及作用。方法:采用RT-PCR法检测CMS患者BMMNCs caspase-3 mRNA表达水平。同时以16例骨科单纯骨折患者为对照。结果:①CMS组caspase-3 mRNA相对表达水平为0.19±0.05,明显低于对照组的0.47±0.10(P<0.01)。②CMS组caspase-3 mRNA表达水平与血红蛋白浓度呈负相关(r=-0.96,P<0.01)。结论:CMS患者BMMNCs中caspase-3表达降低,提示CMS患者BMMNCs凋亡下调,caspase-3参与CMS骨髓造血细胞凋亡异常机理。Objective:To explore the changes of caspase-3 expressions of bone marrow cells in patients with chronic mountain sickness(CMS).Methods:The 18 CMS patients and 16 controls were studied in this study.The levels of caspase-3 mRNA in bone marrow mononuclear cells(BMMNCs) of CMS patients and controls were determined by RT-PCR.Results:①The caspase-3 mRNA of BMMNCs in patients with CMS was 0.19±0.05,and lower than 0.47±0.10 in controls(P<0.01).②Hemoglobin(Hb) was negatively correlated with caspase-3(r=-0.96,P<0.01) in CMS patients.Conclusion:The results showed that the apoptosis of BMMNCs in CMS patients decreased,and caspase-3 might be involved in machanism of this change of BMMNCs apoptosis.
