经熟地鳖甲煎剂含药血清培养的肾系膜细胞对成骨细胞增殖和分化的影响

摘　　要：目的研究经熟地鳖甲煎剂含药血清培养的肾小球系膜细胞培养上清液对成骨细胞的影响。方法采用细胞培养的方法,将成骨细胞分为正常血清组、系膜上清组、含药系膜上清组,分别在培养24、48、72、120h后应用MTT法检测各组成骨细胞的增殖情况;在72h后应用ELISA法检测上清液碱性磷酸酶(AKP)和骨桥蛋白(OPN)浓度。结果系膜上清组和含药系膜上清组的成骨细胞增殖显著高于正常血清组(P<0.05),而含药系膜上清组又高于系膜上清组(P<0.05)。系膜上清组和含药系膜上清组的AKP浓度显著高于正常血清组(P<0.05),含药系膜上清组又显著高于系膜上清组(P<0.05)。含药系膜上清组的OPN浓度要显著高于正常血清组(P<0.05)。结论肾系膜细胞可促进成骨细胞的增殖及功能,而熟地鳖甲煎剂能促进这一作用。本研究为中医"肾主骨"理论体系提供了新的实验方法及科学依据。Objective To explore the effects of glomerular mesangial cells(MC) cultured by prepared rhizome of rehmannia(PRR) and carapax amydae(CA) medicated serum on osteoblasts.Methods The osteoblasts cultured in vitro were divided into three groups: the normal blood serum group,MC group and medicated MC group.After 24 h,48 h,72 h and 120 h,the proliferation of osteoblasts was tested respectively;72 h later,concentrations of alkaline phosphatase(AKP) and osteopontin(OPN) in the osteoblasts were tested by ELISA.Results The proliferation of osteoblasts was significantly increased in the MC and the medicated MC groups compared with that in the normal blood serum group(P<0.05),and it was significantly higher in the medicated MC group than in the MC group(P<0.05).The protein level of AKP in the osteoblasts was significantly higher in the MC group and the medicated MC group than in the normal blood serum group(P<0.05),and the medicated MC group had a higher level than the normal blood serum group(P<0.05).The protein level of OPN in the osteoblasts in the medicated MC group was higher than that in the normal blood serum group(P<0.05).Conclusion MC can promote the proliferation and function of osteoblasts,and this effectiveness can be enhanced by PRR and CA decoction.Our experiment provides a new experimentation for the kidney being in charge of bone,one theory of Traditional Chinese Medicine.
