头孢噻肟对人外周血淋巴细胞钙信号转导作用的研究

摘  要:以人外周血淋巴细胞为模型,利用稳态荧光法研究该模型在外源药物头孢噻肟刺激下,细胞钙稳态的变化,同时考察了钙-ATP酶活性与钙稳态之间的关系.研究发现,在低浓度组的头孢噻肟钠(0.005g/L)刺激下,淋巴细胞内钙离子浓度略有增加,而胞外的钙离子浓度几乎没有发生变化.当药物浓度继续增加,胞内钙离子浓度逐渐减少,药物浓度与胞内钙离子浓度呈现出一定的剂量-效应关系.不同药物处理组的钙-ATP酶活性与对照组相比均降低,呈现出一定的剂量-效应关系.To observe the relationship of Ca2+ ion concentrations with the changes of Ca2+-ATPase in human lymphocytes treated with cefotaxime(CTX) of different concentrations(0.005 g/L,0.05 g/L,0.5 g/L,1 g/L,2 g/L),free calcium concentrations in human lymphocytes were measured by double wavelength spectrofluorometer with Fura-2/AM as the fluorescence indicator.The results showed that low concentration of CTX(0.005 g/L) lead to the increase of i via phosphatidylinositol transduction pathway.In the other groups(0.05 g/L,0.5 g/L,1 g/L,2 g/L),i in human lymphocytes were reduced by the concentration of CTX increased,hence the concentration of CTX and i showed dose-effect relationship.Meanwhile,the activities of Ca2+-ATPase in human lymphocytes were investigated.Compared with the control group,the activities of Ca2+-ATPase in group with cefotaxime were all declined.The concentration of CTX and activities of Ca2+-ATPase also showed dose-effect relationship.
