摘 要:目的探讨无并发症的2型糖尿病(T2DM)患者外周血单个核细胞(PBMC)中核因子-κB/p65(NF-κB/p65)、核因子-κB抑制蛋白α(IκB-α)表达水平的变化及其与氧化应激、糖代谢等因素的关系。方法以无并发症T2DM患者30例、非糖尿病对照(NDM)30例为研究对象,测定其PBMC中NF-κB/p65、IκB-α蛋白表达水平,血清丙二醛(MDA)、超氧化物歧化酶(SOD)及糖化血红蛋白(GHb)等指标变化。结果与NDM组相比,T2DM患者NF-κB/p65表达升高;MDA水平升高,SOD活性降低。NF-κB/p65表达与MDA、GHb、稳态模型胰岛素抵抗指数呈正相关,与SOD呈负相关。T2DM患者与NDM间IκB-α蛋白表达无统计学差异。结论无并发症的T2DM患者PBMC中NF-κB/p65表达升高,其表达水平与氧化应激、血糖控制水平相关。Objective To investigate the changed expression of nuclear factor kappa B(NF-κB) in peripheral blood mononuclear cells(PBMCs) of type 2 diabetes patients without vascular complications and its relationship to oxidative stress and glucose metabolism.Methods The study subjects comprised 30 diabetes patients with type 2 diabetes(T2DM) without vascular complications and 30 non-diabetic controls(NDM).The content of malondiadehyde(MDA) and the activity of superoxide dismutase(SOD) in plasma were detected by chemical colorimetric method.The protein levels of NFκB-p65 and inhibitor kappa B-α(IκB-α) in PBMCs in T2DM patients were measured by Western blot.Other laboratory data including glycosylated hemoglobin(GHb),total cholesterol and triglyceride were evaluated.Results The protein level of NF-κB/p65 was significantly increased in the T2DM patients.Compared to NDM controls,these patients also had significantly higher concentration of MDA and lower SOD activities.The protein level of NF-κB/p65 was positively associated with MDA concentration and GHb.However,there was no significant difference in IκB-α protein level between the T2DM patients and NDM controls.Conclusion Type 2 diabetes patients without vascular complications have an increased NF-κB/p65 expression,which is associated with oxidative stress and the level of glucose control.
