摘 要:目的探讨大鼠颌下腺细胞中是否表达降钙素(CT),为进一步证明颌下腺与甲状腺C细胞具有同源性提供依据。方法采用细胞培养、免疫细胞化学SP法以及原位杂交法进行降钙素的细胞定位及基因定位,并用酶联免疫法(ELISA)测定培养液中降钙素的含量。结果大鼠颌下腺上皮细胞呈CT免疫反应阳性,阳性物质分布于胞质,胞核呈阴性反应。上述细胞同样含有CT mRNA杂交信号,信号物质亦分布于胞质内,胞核呈阴性反应。ELISA法测定大鼠颌下腺细胞培养液中降钙素的含量明显高于对照组。结论大鼠颌下腺上皮细胞能分泌CT,提示大鼠颌下腺上皮细胞可能与甲状腺C细胞具有同源性。Objective To study the expression of calcitonin(CT) in the submandibular gland cell of rats so as to provide further evidence for proving the homology of the submandibular gland cell and thyroid C cell in rats. Methods Cell and gene location of calcitonin was made by cell culture,immunocytochemistry SP methods and in situ hybridization;ELISA was used to detect calcitonin in the culturing supernatant. Results The submandibular gland cell of rats showed CT positive immunoreactivity,which was located in the cytoplasm with negative nuclei.CT mRNA hybridization signals were distributed in the cytoplasm with negative nuclei.The concentration of calcitonin in the culturing supernatant was significantly higher than that of the control group. Conclusion The submandibular gland cell of rats can produce CT,suggesting that there is homology between the submandibular gland cell and thyroid C cell in rats.
