摘  要:DMRT/Dmrt(Doublesexand Mab-3Related Transcription factor)是一类含有锌指样、富含半胱氨酸、能和特异DNA序列结合的DM结构域的转录因子.目前在脊椎动物中已发现8类DMRT/Dmrt基因,其中大部分成员在性腺中表达,表明这些基因可能参与了动物的性别决定与分化,此外部分DMRT/Dmrt成员还参与了体节和神经的形成等胚胎发育的过程,因此Dmrt基因家族是一类与发育相关的重要调控基因.通过RT-PCR方法成功克隆到了热带爪蟾胚胎Dmrt3基因的全长编码区,并进一步通过整体原位杂交的方法发现Dmrt3在热带爪蟾早期胚胎发育过程中主要在嗅觉原基、端脑、侧线原基、神经管等部位特异性表达.其在嗅觉原基、端脑与Dmrt4的重叠表达表明Dmrt3和Dmrt4在爪蟾早期胚胎发育的嗅觉神经形成过程中可能存在功能冗余.DMRT/Dmrt(D-oublesex and M-ab-3 R-elated T-ranscription factor) genes are characterized by a unique zinc finger like DNA-binding motif known as the DM domain.So far,eight different Dmrt members have been identified in vertebrates,and most of the members have been implicated in sexual determination and differentiation since their specific expression in gonads.However,some Dmrt genes also involve in other developmental processes such as the development of somites and olfactory neurogenesis.Thus Dmrt genes consist of an important developmental regulator.Herein,the full length cDNA of Dmrt3 was cloned by RT-PCR from X.tropicalis embryos.Whole-mount in situ hybridization(WISH) revealed that the spatial expression of Dmrt3 was restricted in the olfactory placode,lateral line placode,neural tube and telecephanlon.The overlapping expressions in olfactory placode and telecephalon indicate that Dmrt3 might participate in olfactory neurogenesis like Dmrt4 during Xenopus early embryogenesis.
