摘  要:目的:星点设计—效应面法优选川麦冬总黄酮的提取工艺。方法:以乙醇浓度、液料比、提取时间为自变量,以川麦冬总黄酮含量得率为因变量,通过对自变量各水平的二项式拟合,用效应面法选取最佳工艺,并进行预测分析。结果:确定的最佳工艺为85%乙醇、液料比36:1、温度80℃、提取3h,提取次数1次。提取预测值与验证值偏差为3.76%。二项式拟合复相关系数平方r=0.9610。结论:星点设计-效应面法优化川麦冬的总黄酮提取工艺,精密度高,方法简便,可预测性较好。Objective:To optimize the extraction process of total flavonoids from ophiopogonis radix by central composite design-response surface method.Method:Ethanol concentration,solvent ratio and extraction time were independent variables.Extraction rate of total flavonoids in ophiopogonis radix was dependent variable.No-linear mathematic models were used to estimate the relationship between the independent and the dependent variables.Response surface methodology was used to optimize extraction process.Predictions were carried out by comparing the observed and predicted values.Result:Optimal conditions of extraction process were as following:refluxed one time at 80℃ with 36 ratio of 85% ethanol for 3 hours.The bias between the observed and the predicted values of extraction quantity was 3.76%.Regression coefficient of binomial fitting complex model was as high as 0.9610.Conclusion:The optimum model is simple,precise and highly predictive.
