摘 要:目的:采用高效液相技术(HPLC)检测实验大鼠血清中穿心莲内酯及脱水穿心莲内酯的含量。方法:穿心莲水粗提取物分别给予实验大鼠连续灌胃7d和12d后取血清,采用高效液相仪分析大鼠血清中的穿心莲内酯和脱水穿心莲内酯,并明确其在体内的含量。HPLC条件:色谱柱Waters Symmetry Shield C18柱(4.6×250mm,5μm);流动相甲醇∶水(6.5∶3.5);紫外检测波长240nm。结果:大鼠血清用HPLC检测显示7d和12d大鼠血清中穿心莲内酯含量分别为3.12×10-6和1.512×10-6g/mL,脱水穿心莲内酯含量均较少。结论:在大鼠血清中均可检测到穿心莲内酯及脱水穿心莲内酯,但检测到的脱水穿心莲内酯较少,可能是因为脱水穿心莲内酯难溶于水中,使水煎液中含量有限;也可能是脱水穿心莲内酯在体内很快转化为了其他代谢产物。Objective: High performance liquid chromatography (HPLC) was used to detect the contents of andrographolide and dehydroandrographolide in rat serum. Method:After continuously intragastric administration of water extract of Andrographis Herba to rats for 7 or 12 days, the rat serum was collected and analyzed by HPLC to detect the chemical compositions and the contents. The analysis was carried out in Waters Symmetry Shield C18 column (4.6×250 nm, 5 μm). The mobile phase was methanol:water (6.5:3.5) and the detection wavelength was set at 240 nm. Result:The contents of andrographolide in rat serum were 3.12×10-6 and 1.512×10-6 g﹒mL-1, respectively. And the content of dehydroandrographolide was few in rat serum. Conclusion:Andrographolide and dehydroandrographolide can be detected in rat serum. The reason for few dehydroandrographolide in rat serum may be related to the low solubility in water, or the rapid transformation to metabolite in vivo.
