运用改良的单侧肾切多次注射OX7抗体方法建立慢性系膜增殖性肾炎模型

摘　　要：目的运用改良的单侧肾切多次注射OX7抗体方法建立慢性系膜增殖性肾炎模型。方法雄性Wistar大鼠行单侧肾切除后随机分为单纯肾切组、单侧肾切单次注射OX7抗体组和单侧肾切多次注射OX7抗体组。单侧肾切单次注射OX7抗体组在单侧肾切除后3天尾静脉单次注射OX7抗体(100mg/kg),单侧肾切多次注射OX7抗体组在单侧肾切除后3天开始尾静脉多次注射OX7抗体(每次100mg/kg,1次/周,共4次),观察大鼠的一般情况、体质量、血压、尿蛋白、肾功能以及肾脏病理改变。结果单侧肾切单次注射OX7抗体和单侧肾切多次注射OX7抗体的方法均可以成功建立慢性系膜增殖性肾炎模型,多次注射组在第8周时出现肾小球节段性硬化,肾小管灶状萎缩;而单次注射组在第16周时才出现上述病理改变。多次注射组肾小球及肾间质小管间质积分明显高于单次注射组(P<0.05)。结论与国外常用的单侧肾切单次注射OX7抗体方法相比,用单侧肾切多次注射OX7抗体的方法,可以更快地建立慢性系膜增殖性肾炎大鼠模型。Objective To set up an experimental progressive mesangial proliferative glomerulonephritis model by one side nephrectomy combined with multiple injections of antibody to OX7.Methods Male Wistar rats were subject to one side nephrectomy,and the survival rats were randomly assigned to 3 groups:rats with single injection of antibody to OX7,rats with multiple injections of the antibody,and rats without the injections as controls.Antibody to OX7 was injected through caudal vein 3 days after nephrectomy,once for the single injection of antibody to OX7 group and once a week for 4 times for the multiple injections of antibody to OX7 group.General status,body weight,systolic blood pressure,proteinuria,serum creatinine,glomerular and tubulointerstitial histological damage scores were observed in the rats.Results The progressive mesangial proliferative glomerulonephritis model was set up by single injection or multiple injections of antibody to OX7 in one side nephrectomy rats.Segmented glomerular sclerosis and renal tubular focal atrophy were observed in multiple injections group at week 8,and the same pathological changes were observed in single injection group at week 16.Conclusion Rat model of progressive mesangial proliferative glomerulonephritis was set up in one side nephrectomy rats treated in a shorter period of time with multiple injections of antibody to OX7 and in longer time with single injection of antibody to OX7.
