摘  要:目的探讨北冬虫夏草腺苷的提取方法和检测技术。方法用微波萃取装置提取腺苷,考查水、甲醇、乙醇三种溶剂的提取效果,并进一步考察了固液比、萃取温度、萃取时间的影响,并对HPLC法测定提取液的腺苷含量的方法进行了方法实验研究。结果以水为提取溶剂,温度50℃,固液比为1∶20,提取时间15 min时提取效率0.233%。选用C18柱(4.6 mm×250,5μm),流动相:水∶甲醇=85∶15,流速0.8 mL/min,检测波长270 nm条件下,腺苷准确度符合方法学要求。结论采用微波萃取法提取北冬虫夏草中的腺苷,利用HPLC测定其含量的方法具有实用性并有推广应用价值。Objective To study extraction method and detection technology of adenosine from Cordyceps.Methods Adenosine was extracted by microwave sample to examine the effects of extraction of three different solvents:water,methanol and alcohol,and effects of solid—to—liquid ratio,extraction temperature and time,the extracted amount was examined by HPLC.Results The extraction efficiency could reach 0.233% when the solvent was water,temperature was 50℃,solid—to—liquid ratio was 1∶20,extraction time was 15 mins.Adenosine accuracy met the requirements of methodology under the following conditions:C18 column(4.6 mm×250,5 μm),mobile phase:water∶methanol=85∶15,flow rate 0.8 mL/min,detection wavelength:270 nm.Conclusion The method used to extract adenosine from Cordyceps and to determine the content by HPLC was worth spreading.
