鸡抗CHO细胞蛋白的IgY消长规律及其制备的试验研究

摘　　要：采用五种免疫程序分别免疫来亨鸡以制备抗体;ELISA检测不同免疫方案鸡抗体水平变化规律。结果显示,不同免疫程序接种来亨鸡后抗体产生水平不同。末针免疫28d后,4针低剂量肌肉免疫组抗体水平显著低于4针高剂量肌肉免疫组(P<0.01);3针低剂量肌肉免疫组抗体水平显著低于3针高剂量肌肉免疫组(P<0.01);肌肉免疫组抗体水平低于皮下免疫组(P<0.05)。所有组均在末针免疫20d后出现抗体高峰,抗体水平可持续30d以上;SDS-PAGE结果显示纯化IgY纯度可达99%,抗体ELISA效价大于1∶105。To explore the regularity of chicken IgY against CHO cell protein and its immunization efforts,and then to prepare chicken IgY of high titer.Leghorns were divided into five groups and were used five different immunization procedures respectively to prepare antibodies.ELISA was applied to detect the regularity of chicken IgY against CHO cell protein.The antibody levels of Leghorns immunized with five different immunization programs are different.Antibody level of quartic low dose intramuscular injection group is significantly lower than that of the quartic high-dose intramuscular injection group(P<0.01);Antibody level of triple low-dose intramuscular injection group is significantly lower than that of the triple high-dose intramuscular injection group(P<0.01);Antibody level of intramuscular injection group is lower than that of the hypodermic injection group(P<0.05).All groups appear antibody peaks at the 20th day after the last immunization.SDS-PAGE analysis showed that the purity of purified IgY is 99% and its ELISA titer is greater than 1∶ 105.From the 28th day after the end immunization,the antibody level of low-dose intramuscular injection group is lower than that of the high-dose intramuscular injection group.And of the same dose,the antibody titer of the intramuscular injection group is lower than that of hypodermic injection group.Chicken IgY of high titer against CHO cell soluble protein has been successfully prepared.
