高效液相色谱法测定活肾丸中丹参酮ⅡA的含量

摘　　要：目的活肾丸现行质量标准只采用薄层色谱法进行定性鉴别。文中建立测定活肾丸中丹参酮ⅡA含量的高效液相色谱法。方法采用Lichrospher C18色谱柱(4.6 mm×250 mm,5μm),甲醇-水(80∶20)为流动相,流速1.0 ml/min,检测波长为267 nm。结果丹参酮ⅡA的浓度在5.015～80.24μg/ml范围内具有良好的线性关系,r=0.9999;平均回收率为98.92%,RSD为1.92%(n=9),活肾丸中丹参酮ⅡA的含量最低应不少于0.08 mg/g。结论本法操作简单,检测结果准确,重复性好,可作为该制剂的质量控制标准。Objective Thin layer chromatography(TLC) is normally used to identify the quality of Huoshen Pills.This article introduces the employment of high performance liquid chromatography(HPLC) in measuring the content of tanshinone ⅡA in Huoshen Pills.Methods To determine the content of tanshinone ⅡA in Huoshen Pills,we performed HPLC on Lichrospher C18(4.6mm×250mm,5μm) with methanol-water(80∶20) as the mobile phase,the flow rate at 1.0ml/min and the detecting wavelength of 267nm.Results The linear range of tanshinoneⅡA was 5.015-80.24μg/ml,r=0.9999.The average rate of recovery was 98.92%,and the RSD was 1.92%(n=9).The content of tanshinone ⅡA in Huoshen Pills was at least 0.08mg/g.Conclusion HPLC is simple,accurate and reproducible,which can be used for the quality control of Huoshen Pills.
