摘 要:目的:探讨山药多糖对小鼠镉染毒所致急性肝损伤的预防作用及其机制。方法:昆明种小鼠50只随机分为5组,其中对照组和模型组常规饲料,饮自来水;山药多糖低、中、高剂量组:按山药多糖计算每kg常规饲料中分别加2.5、5.0、10.0 g山药多糖,自由饮水,饲养7 d。第7天除对照组腹腔注射等体积的0.9%氧化钠溶液外,其余各组均按3.0 mg/kg腹腔注射CdCl2溶液。24 h取血测量血清ALT和AST。取肝脏称重计算肝体指数(肝质量/体质量),制备肝匀浆测定SOD、GSH-Px活性及MDA、NO的含量。结果:山药多糖能降低肝体指数,降低血清ALT、AST活性和MDA、NO含量,提高肝脏SOD、GSH-Px活性和GSH的含量,与模型组比较有统计学意义(P<0.05,P<0.01)。结论:山药多糖可通过拮抗镉染毒所致的氧化应激反应,对肝损伤发挥保护作用。Objective To investigate the prevention of Chinese yam polysaccharides on cadmium-induced liver injury in mice and its mechanism.Methods Fifty Kunming mice were randomly divided into five groups,the mice in control group and model group given normal diet and tap water,the mice in other three Chinese yam polysaccharides groups given conventional food and Chinese yam polysaccharide with a dose of 2.5,5.0,10.0 g every kilogram body weight for seven days,respectively.On 7th day,all groups were intraperitoneally injected CdCl2 with a dose of 3.0 mg/kg,except that the control group was intraperitoneally injected physiological saline with equal volume.The serum levels of ALT and AST were determined and the hepatosomatic indexes(liver weight/body weight) were calculated after 24 hours,and then liver homogenate was prepared to detect the activity of SOD and GSH-Px and the contents of MDA and NO.Results Chinese yam polysaccharide significantly decreased the hepatosomatic indexes,the activities of ALT and AST in serum,meanwhile,it significantly increased the activities of SOD and GSH-Px and the contents of GSH in liver(P<0.05,P<0.01).Conclusion Chinese yam polysaccharide can pretect the cadmium-induced liver injury in mice by antagonizing Cd toxicity and oxidative stress.
