摘  要:目的对白纹伊蚊雌蚊各个组织器官的Wolbachia和WO噬菌体进行时空定量分析,并从量上寻找Wolbachia、WO噬菌体和宿主三者之间的相关性,为进一步揭示这三者之间相互作用的分子机制奠定基础。方法分别于第1、5、10、15、20、30、40d各取雌蚊10只,麻醉后于解剖镜下解剖蚊虫的头部、卵巢、中肠、马氏管、脂肪体和胸部,各自提取其基因组DNA,以蚊虫基因rpS7为内参,Q-PCR检测各个组织中的Wolbachia和WO噬菌体的在白纹伊蚊雌蚊体内的平均相对含量和时空动态变化,对Wolbachia和WO噬菌体的拷贝数进行量化关系转化。结果 Q-PCR结果显示Wolbachia和WO噬菌体在蚊虫的卵巢中含量最大,其次是脂肪体、马氏管和头部,而在中肠和胸部的含量最小。发现在蚊虫d龄前30d中,wAlbB与WO噬菌体的动态变化趋势基本一致,且从拷贝数量上二者存在着线性关系。结论白纹伊蚊雌蚊的各个组织均存在wAlbA和wAlbB的双重感染和WO噬菌体的侵染,且这三者在卵巢中的含量远大于其他组织部位。WO噬菌体主要侵染的对象可能是wAlbB,而不是wAlbA。By measuring the relative density of Wolbachia and phage WO in different Aedes albopictus organs using QPCR,we try to find the quanity relationships among Wolbachia,phage WO and its host,which is basic for the further studies of moleculer mechandisms among them.Five female mosquitoes were collected at 1,5,10,15,20,30,40day respectively,and different mosquito organs were dissected under anatomy mirror.The DNA was extracted from the head,ovary,midgut,malpighian tubules,fat body and thorax.Q-PCR was used to measure the relative density and dynamic change of Wolbachia and phage WO in the extracted organs.Both of their copies were measured in order to establish the quanity relationships between them.Q-PCR showed that the relative density of Wolbachia and phage WO were highest in ovary,and then were fat body,malpighian tubules and head,midgut and thorax were at least.We found that the variation tendency of wAlbB and phage WO was similar in the front-life cycle of mosquito(30day),who have linear relationships in quanity.Different Aedes albopictus organs were superinfected with Wolbachia and phage WO.The relative density of Wolbachia and phage WO in ovary were far higher than that in other organs.It might be wAlbBbut not wAlbAinfected with phage WO.
