摘 要:[目的]探讨野山杏提取物(WAE)对高脂血症大鼠脂代谢的影响。[方法]在建立大鼠高脂血症模型的基础上,观察不同剂量WAE对大鼠血清脂质水平以及血清谷丙转氨酶(ALT)和谷草转氨酶(AST)、肝脏中的脂肪代谢酶和MDA的活性以及对肝脏病理学变化的影响。[结果]WAE对高血脂大鼠血清中的TC、TG与LDL-C的水平与模型组相比有明显的降低,0.2 g/(kg·d)剂量的WAE分别降低到4.61±0.58、0.44±0.06、4.34±2.57 mmol/L;0.4 g/(kg·d)剂量的WAE分别降低到3.75±0.39、0.42±0.07、2.53±0.8 mmol/L;0.4 g/(kg·d)剂量的WAE对HDL-C的水平与模型组相比有明显的升高,升高到1.19±0.34mmol/L;对ALT、AST活性与模型组相比有明显降低,0.2g/kg剂量的WAE分别降低到9.94±2.34、32.04±5.86 U/L,0.4 g/(kg·d)剂量的WAE分别降低到10.49±1.75、26.59±3.86 U/L;WAE对高血脂大鼠肝脏在MDA含量与模型组相比有明显降低,分别降低到0.2 g/kg剂量的WAE为1.91±0.45 U/mgprot,0.4 g/(kg·d)剂量的WAE为1.72±0.57 U/mgprot;对LPL的活性与模型组相比有明显的升高,0.2 g/kg剂量的WAE升高到13.32±2.57 U/mgprot,0.4 g/(kg·d)剂量的WAE升高到11.92±2.77 U/mgprot;0.4 g/(kg·d)剂量的WAE对HL的活性与模型组相比明显的升高,升高到11.02±1.57 U/mgprot;对LPS的活性与模型组相比有显著的降低作用,0.2剂量和0.4 g/(kg·d)剂量的WAE分别降低为4.40±1.68和3.51±1.61 U/mgprot;组织切片镜检可见肝脂肪变性程度减轻,肝细胞结构接近正常水平。[结论]WAE能降低高脂血症大鼠的血脂水平,有抗动脉粥样硬化形成的作用,在一定程度上预防脂肪在肝脏中的沉积,且作用效果呈现一定的剂量依赖性。[Objective]The purpose of this research was to investigate the effect of wild apricot extracts (WAE) on lipid metabolism of hyperlipidemia rats.[Method]On the basis of establishing hyperlipidemia rat model,effect of WAE on the serum lipids level,the activities of ALT and AST in serum,lipid metabolite enzyme in liver,the activities of MDA,and pathology change of liver were investigated.[Result]WAE could obviously decrease the content of TC,TG,LDL - C and the activities of ALT and AST and increase the content of HDL - C of serum in hyperlipidemia rats drenched with high fat suspension.WAE also lowered level of MDA in liver tissue.The degrees of hepatic steatosis:n WAE groups were lighter than those in hyperlipidemia model group,especially in the high dose group,which was close to normal essentially.WAE plays an important role in enhancing the LPL and HL activity and inhibiting LPS.[Conclusion]WAE indeed had the function of reducing blood lipid of hyperlipidemia rats in addition to the counteraction atherosclerosis.To some extent,WAE also had good prevention to fatty deposition in liver.Moreover,treatment of WAE showed a certain relationship with dosage.
