急性内毒素损伤对奶山羊肝脏营养代谢的影响

摘　　要：本文旨在研究急性内毒素(LPS)损伤对奶山羊肝脏营养代谢的影响,初步探讨其作用机制。选取年龄、体重相近的关中奶山羊母羊18只,随机分为3组,分别作为对照组(CTL组)、试验Ⅰ组(TⅠ组)和试验Ⅱ组(TⅡ组),每组6只。TⅠ组、TⅡ组奶山羊分别按照100和200μg/kg BW的剂量从颈静脉注射LPS注射液30 mL,CTL组奶山羊注射相同体积的生理盐水。在注射后0、1、4、8、12、24 h从奶山羊的颈静脉采血,制备血浆以检测肝脏代谢功能及营养代谢相关指标。结果表明:注射LPS后肝脏代谢功能受到不同程度地损伤。血浆谷丙转氨酶活性平均值,TⅠ组显著高于CTL组(P<0.05),TⅡ组显著低于CTL组(P<0.05);血浆谷草转氨酶活性平均值,TⅠ组显著高于TⅡ组和CTL组(P<0.05),TⅡ组和CTL组之间差异不显著(P>0.05)。随着时间推移,血浆葡萄糖含量在CTL组基本稳定,在TⅠ组和TⅡ组出现较大波动,但其平均值各组间无显著差异(P>0.05)。血浆总蛋白含量平均值TⅠ组、TⅡ组显著低于CTL组(P<0.05)。随着时间的推移,血浆白蛋白含量在CTL组基本稳定,在TⅠ组和TⅡ组逐渐降低,其平均值各组间无显著差异(P>0.05)。血浆尿素氮、甘油三酯、低密度脂蛋白、高密度脂蛋白和胆固醇含量平均值均表现为TⅠ组>TⅡ组>CTL组,除血浆低密度脂蛋白外,其他各指标均表现为TⅠ组、TⅡ组显著高于CTL组(P<0.05)。此外,TⅠ组血浆高密度脂蛋白和胆固醇含量平均值显著高于TⅡ组(P<0.05)。由此得出,LPS可引起肝脏损伤,进而对奶山羊肝脏营养代谢造成影响;LPS对奶山羊肝脏营养代谢的影响与LPS的剂量有关。This experiment was conducted to investigate the effects of acute lipopolysaccharide(LPS) injury on liver nutrition metabolism in dairy goats,and to explore its possible mechanism.Eighteen female dairy goats with the similar body weight and age were selected and randomly divided into 3 groups which were designed as control group(CTL group),experimental Ⅰ group(TⅠ group) and experimental Ⅱ group(TⅠ group).The goats in the TⅠ and TⅠ groups were injected 100 and 200 μg/kg BW LPS of 30 mL via jugular vein,respectively,while the goats in the CTL group were injected the same volume of normal saline.The blood was collected via jugular vein at 0,1,4,8,12 and 24 h after injection to make plasma to detect the related indices of liver metabolism function and nutrition metabolism.The results showed as follows: the liver metabolism function was injured in different degree after injection of LPS.Plasma alanine aminotransferase(ALT) activity,which in CTL group was significantly lower than that in TⅠ group(P<0.05) and significantly higher than that in TⅡ group(P<0.05).For plasma aspartate aminotransferase(AST) activity,TⅠ group was significantly higher than CTL group and TⅡ group(P<0.05),but no significant difference between CTL group and TⅡ group(P>0.05).With the passage of time,plasma glucose(GLU) content in CTL group was basically stable,and that in TⅠ and TⅡ groups had a large fluctuation,but no significant difference in its mean among all groups(P>0.05).Mean of plasma total protein(TP) content in TⅠ and TⅡ groups was significantly lower than that in CTL group(P<0.05).With the passage of time,plasma albumin(ALB) content in CTL group was basically stable,and that in TⅠ and TⅡ groups presented a gradually decreasing trend,but no significant difference in its mean among all groups(P>0.05).Means of contents of plasma urea nitrogen(UN),triglyceride(TG),high density lipoprotein(HDL),low density lipoprotein(LDL) and cholesterol(CHOL) were presented as TⅠ group>TⅡ group>CTL group,and above indi
