摘 要:目的探讨改良式筛网法在分离子宫内膜腺上皮细胞及间质细胞中的应用价值。方法通过高浓度胶原酶结合不同浓度、时间消化、过滤及贴壁纯化等技术,应用筛网分离法培养各15例子宫内膜腺上皮和间质细胞,并传代。结果改良式筛网分离法培养中的15例标本中,腺上皮细胞和间质细胞成功率分别为93.3%和93.3%;分离出的腺上皮细胞及间质细胞可体外培养,经分离纯化后腺上皮细胞纯度>90%,间质细胞纯度>98%;两种细胞经传代后间质细胞成功率92.9%;腺上皮细胞传代培养后活力下降,成功率14.3%。结论通过改良式筛网分离法能获得纯度较高的腺上皮细胞与间质细胞;间质细胞可行传代培养;腺上皮细胞传代培养后活力下降,不宜传代。Objective To explore the value of improved screen isolated and purified endometrial cells.Methods By high concentrations of collagenase,different concentrations,time to digest,filter and adherence purification technology,sieve separation method ware used for culture and subculture 15 endometrial glandular epithelium and stromal cells.Results 93.3 % well-grown epithelial cells were obtained and 93.3% stromal cells via method of improved screen separation,Isolated glandular epithelium epithelial cells and stromal cells can be cultured in vitro.After separation and purification,the glandular epithelial cell purity was more than 90% stromal cell purity was more than 98%.Interstitial cells of the success rate of 92.9%;glandular epithelial cell subculture decreased vigor,the success rate of 14.3%.Conclusion Through the improved screen separation can train mesenchymal and epithelial cells of high purity.
