摘 要:采用显微镜计数法、荧光分光光度法、叶绿素a含量测定法、可见分光光度法对利玛Coolia monotis指数生长期不同浓度的培养液进行细胞密度测定.结果表明后3种细胞计数方法与显微镜计数法相比较,均有较好的线性关系;同时对不同生长期的Coolia monotis培养液进行测定的结果表明,叶绿素a含量测定法与显微镜计数法得到的生长曲线一致性最好,最能真实反映Coolia monotis的生长状态,其次是分光光度法,最差的是荧光测定法.比较分析发现,叶绿素a含量测定法适用于不同细胞密度范围的测定,具有简便、计数准确、适于批量操作的优点,可确定为便捷、有效、可行的Coolia monotis细胞计数法.In this study four cell-counting methods of Microscope,Fluorescence spectrophotography,Visible spectrophotography and Chla containing measurement were used to measure the amount of the exponential phase cell of Coolia monotis.The insightful comparison on the interrelations of the last three mentioned cell counting methods was made with the method of Microscope,the results revealed the sound 1inear interrelations among them.Different growth phase of Coolia monotis were simultaneously measured,and then growth curves were drew basis on the data from different methods.The results showed the best concordance between Microscope and Chla containing measurement methods,and then Visible spectrophotography,the worst was Fluorescence spectrophotography.Particularly,for Chla containing measurement method,the operating procedures are simple,accurate,suitable for wide cell-density ranges and batch measurements.Therefore,Chla containing measurement is convenient,high-efficiency and reliable approach for cell counting of Coolia monotis.
