摘 要:目的探讨吸入麻醉药七氟烷是否能在小鼠肝及脑组织中进行生物转化,并对其代谢产物六氟异丙醇(HFIP)进行测定。方法在七氟烷麻醉下,取小鼠部分肝及全部脑组织,将其匀浆后于密闭空针中分别加入0.5%、1%及3%七氟烷于37℃恒温箱中孵育6h,分别于0、1、2、4、6h测定气相中HFIP浓度。结果在对小鼠肝脏组织匀浆的孵育中,3种七氟烷孵育浓度均检测出HFIP,且以0.5%七氟烷孵育时HFIP含量最高。在对小鼠脑组织匀浆的孵育中,当七氟烷浓度为0.5%及1%时检测出HFIP,且前者高于后者,但当七氟烷孵育浓度为3%时,未能检出HFIP。结论七氟烷能在小鼠肝及脑组织中代谢分解成HFIP,但随着七氟烷孵化浓度的增加,HFIP的生成却减少。Objective To investigate whether sevoflurane can be metabolized in mice's liver and brain tissues,and determine the contents of its metabolic product Hexafluoro-isopropanol(HFIP).Methods The liver and brain tissues of mice were obtained under inhaled sevoflurane anesthesia.Liver and brain homogenate were incubated for 6hr with 0.5%,1% and 3% sevoflurane vapor under 37℃,respectively.HFIP was determined at 0,1,2,4 and 6hr during incubation.Results HFIP was found in all liver and brain tissue homogenate with different incubation sevoflurane concentrations(except for brain tissue ####
