PP_(333)调控黄瓜子叶节培养物的花芽形态分化

摘  要:应用离体黄瓜子叶节花芽分化实验系统,研究了PP333在黄瓜花芽分化进程中的作用.组织培养结果显示,0.3mg·L-1PP333处理显著促进离体黄瓜子叶节培养物的花芽分化,直接花形成率由对照的16.86%升高至36.02%,直接花芽/总芽的比值达到最高,约为对照的1.7倍;营养芽形成率和营养芽/总芽的比值均降至最低,分别为对照的72%和59%.组织切片结果显示,0.3mg·L-1PP333处理明显提高黄瓜子叶节培养物的花芽原基百分率和花芽原基/总原基的百分率,增幅分别为42%和50%,而营养芽原基百分率则较对照降低约47%.实验结果发现,适宜浓度的PP333处理可以促进培养物所生成原基向花属性方向发育,是PP333处理促进黄瓜子叶节培养物花芽形成的主要原因.This paper presents a study of the effects of PP333treatment on the initial stage of floral transition of cotyledon node cultures of Cucumis sativus L..The results were as follows: Compared with the control group,the floral transition in the experimental group was increased significantly by the treatment with 0.3 mg·L-1 PP333,during which the formation rate of direct floral buds rose from 16.86% to 36.02%,and the ratio of direct floral buds/total buds reached the highest about 1.7 times as high as the control.In contrast,the formation rate of vegetative buds lowered to 72% of the control,and the ratio of vegetative buds/total buds was lowered to 59% of the control.By histological sectioning,we observed and counted the number of different types of primordia formed.It is obvious that the treatment of 0.3 mg·L-1 PP333markedly increased the percentage of the floral transition of primordia and the ratio of floral primordia/total primordia by 42% and 50% respectively.However,the percentage of vegetative bud primordia was 47% lower than the control.In short,by promoting the transition from primordia generated by cucumber cotyledon node cultures to floral primordia,the treatment with PP333at an appropriate concentration was beneficial to flower initiation.At the end of the paper,some presumed reasons why the treatment of PP333on cotyledon node cultures of cucumber could enhance the floral transition are discussed.
