摘 要:以家鸡血细胞为材料,聚乙二醇(MW=4000)为诱导剂,比较了细胞融合过程中细胞与聚乙二醇混匀的两种方法对融合的效应.用快速混匀器混匀时,细胞融合率、双核融合率与多核融合率比例、融合率稳定性均高于徒手混匀;提示快速混匀器混匀可以替代徒手混匀.测试了聚乙二醇浓度对混匀效果的影响;讨论了混匀器混匀方法在细胞融合中的应用.Two methods for mixing cells with poly(ethylene glycol)(PEG) solution in induced cell fusion are compared when using chicken red blood cells as material and poly(ethylene glycol)(MW=4000) as fusogen.The results shows that the fusion rate,the ratio of two-cells fusion to multi-cells fusion and the stability of fusion rate are all higher when the cells are mixed with PEG by vibration mixer than by freehand operation.This suggests the practicability of substitution of freehand mixing by vibration device mixing.Also the impact of PEG concentration on the mixing condition is tested.The application possibility of vibration device mixing method in PEG induced cell fusion is discussed.
