摘 要:目的比较研究哮喘大鼠模型地塞米松干预撤停不同阶段(撤停前、撤停中、撤停后)合用淫羊藿女贞子的协同抗炎作用。方法卵清白蛋白(ovalbumin,OVA)致敏激发建立大鼠哮喘模型,腹腔注射地塞米松模拟激素干预过程,分别在激素干预不同阶段合用中药淫羊藿女贞子,观察比较药物对激素干预各阶段哮喘模型支气管肺泡灌洗液(branchoalveolar lavage fluids,BALF)涂片嗜酸粒细胞(eosinophils,EOS)计数,BALF中γ-干扰素(interferon-γ,IFN-γ)、白介素(interleukine,IL)-4、IL-5、肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)含量。结果①激素撤停前阶段:淫羊藿女贞子合用激素干预组能下调哮喘模型EOS数量和IL-5、IL-4、TNF-α含量,上调IFN-γ表达,调节Th1/Th2失衡。②激素撤停中阶段:淫羊藿合用女贞子合激素干预Ⅰ、Ⅱ组能显著下调哮喘模型大鼠EOS数量、IL-5水平;淫羊藿女贞子合用激素干预Ⅰ组能显著提高IFN-γ/IL-4比值;淫羊藿女贞子合用激素干预Ⅱ组可显著抑制TNF-α含量。③激素撤停后阶段:激素干预组EOS炎症加重;淫羊藿女贞子合用激素干预Ⅰ、Ⅱ组能下调哮喘模型大鼠EOS数量,淫羊藿女贞子合用激素干预Ⅱ组对IL-5也有显著的抑制作用;淫羊藿女贞子合用激素干预Ⅰ、Ⅱ、Ⅲ组均能降低IL-4含量,升高IFN-γ含量,仅淫羊藿女贞子合激素干预Ⅰ组对IFN-γ/IL-4有显著调节作用,还能显著降低TNF-α含量。结论激素应用可有效地抑制EOS为主的气道炎症。但随着激素用量的撤减及停用,其抗炎作用减弱,气道炎症加重。淫羊藿女贞子在激素撤停早期介入与糖皮质激素的协同增效抗炎作用更优。Objective To observe the effects of the compatibility of Epimedium and Ligustrum lucidum (YN) on airway inflammation of asthmatic in rats with glucocorticoid (GC) intervention at different treatment stage (before withdrawal, during the withdrawal and after withdrawal).Methods Dexamethasone was injected into asthmatic model in rats with the oral gavage of YN. The index of airway inflammation was detected at the same time, including IFN-γ, TNF-α, IL-4, IL-5 comcentrations and EOS number branchoalveolar lavage fluids (BALF).Results Before the stage of withdrawing GC, group of YN and GC can reduce the number of EOS, the levels of IL-4, IL-5, and TNF-α,and increase the level of IFN-γ. During the GC withdrawal stage, group Ⅰ and Ⅱ of YN and GC can reduce the number of EOS and the level of IL-5, especially the effect of group Ⅰ of YN and GC to IL-5. Group Ⅰ of YN and GC can modulate IFN-γ/IL-4, and group Ⅱ of YN and GC can restrain TNF-α. After the GC stop stage, EOS number of model group with GC treatment is exacerbated. Group Ⅰ and Ⅱ of YN and GC can reduce the number of EOS. Group Ⅰ, Ⅱand Ⅲ of YN and GC can reduce the level of IL-4, and increase IFN-γ. Group Ⅰ of YN and GC can regulate IFN-γ/IL-4 and the level of TNF-α significantly. And group Ⅱ of YN and GC restrains IL-5.Conclusions YN have different effects of anti-inflammatory on asthmatic model of rats with the compatibility of GC. It can be infered that the compatibility of YN and GC has the tendency of synergy.
