摘 要:利用水 水溶性有机溶剂体系,研究了来源于恶臭假单胞菌的海因酶催化DL 对羟基苯海因合成N 氨基甲酰 D 对羟基苯甘氨酸,以制备D 对羟基苯甘氨酸,并就培养基的筛选、有机溶剂的选择和有机溶剂质量分数的优化作了探讨.结果发现,适量的有机溶剂可增加底物的溶解度,进而显著增加酶反应速度.培养基以质量分数为1%的甘油、1%的酵母膏、0 2%的K2HPO4·3H2O、0 3%的NaCl、0 05%的MgSO4·7H2O和0 1%的诱导剂为佳;有机溶剂以二甲亚砜为宜,其质量分数以10%左右为佳.在此优化反应条件下,水 水溶性有机溶剂体系中酶反应的速度较水相中提高近4倍.The synthesis of N-carbamyl-D-p-hydroxyphenylglycine for the preparation of D-p-hydroxyphenylglycine from DL-p-hydroxyphenydantoin catalyzed by D-hydantoinase from Pseudomonas putida in water-water soluble organic solvent system was studied. The filtration of culture medium, the effects of organic solvent and the content of organic solvent on the reaction rate were also explored. It was found that the organic solvent can increase the solubility of the substrate and quicken the enzymatic reaction rate remarkably. The better composition of culture medium was 1% glycerol,1% yeast grease,02%K_2HPO_4·3H_2O,03%NaCl,005%MgSO_4·7H_2O,01%inducer. Under the optimum conditions such as 10% dimethylsulfoxide as the organic solvent, the enzymatic reaction rate in water/water-soluble organic solvent system was about four times higher than that in aqueous system.
