补肾阳复方激活肾上腺皮质类固醇合成酶的研究

摘　　要：目的研究补肾阳复方升高肾阳虚模型大鼠血浆皮质酮的机制。方法 30只SD大鼠分为3组:对照组、模型组、自拟补肾阳复方组(复方组)。模型组及复方组采用皮质酮10mg/kg皮下注射14d造模;同时复方组采用自拟补肾阳复方按292mg/(kg·d)剂量灌胃;对照组、模型组采用等体积生理盐水灌胃。采用ELISA法检测血浆皮质酮和促肾上腺皮质激素(ACTH)浓度;取肾上腺,进行全基因组表达谱芯片检测及分析,筛选各组差异表达基因;进一步采用Westernblot确认芯片分析结果。结果与模型组比较,复方组血浆皮质酮及ACTH水平显著升高(P<0.05),肾上腺质量显著增加(P<0.05),差异均有统计学意义。基因表达谱分析显示自拟补肾阳复方能提高多种类固醇合成酶如HSD11B1、CYP7A1的表达;Westernblot亦显示HSD11B1蛋白表达升高。结论提高类固醇合成酶的表达可能是补肾阳复方升高肾阳虚证模型血浆皮质酮水平的机制之一。Objective To investigate the effects and mechanisms of Kidney Yang tonifying formula on the expression of adrenal cortical steroid synthetase. Methods Thirty SD rats were divided into control group, model group, Kidney Yang tonifying formula group(compound group). Model group and compound group were injected subcutaneously with corticosterone (10 mg/kg). At the same time, compound group was intragastricly administrated with Kidney Yang tonifying formula[292 mg/(kg·d)],and the other two groups were intragastricly administrated with normal saline. The levels of plasma adrenocorticotrophin (ACTH), corticosterone were detected by ELISA. Expression profiling of whole genome was analyzed on adrenal gland sample to find differently expressed genes. Genechip research results were verified by western blot. Results The levels of ACTH and corticosterone and the adrenal gland wight in compound group were significantly increased than those in model group.The expression of cortical steroid synthetase such as HSD11B1,CYP7A1 was improved in compound group by analysis of gene expression profiling. Conclusions Promoting the expression of steroid synthetase in adrenal gland may be one of underlying mechanisms of Kidney Yang tonifying drug in increasing plasma level of corticosterone.
