摘 要:目的观察重组日本血吸虫磷酸丙糖转移酶(rSj-TPM)在小鼠抗血吸虫感染中的免疫保护作用,寻找新的血吸虫疫苗候选分子。方法以rSj-TPM每鼠50μg/100μl、100μg/100μl和200μg/100μl分别免疫BALB/c小鼠,同时设生理盐水对照组,尾蚴攻击模型小鼠,计算减虫率、减卵率,观察它们的抗血吸虫感染作用;病例切片观察肝脏组织中肉芽肿的变化。结果用重组Sj-TPM免疫BALB/c小鼠,1、2及3组诱导的减虫率分别为27.1%、32.1%和35.2%;1、2及3组诱导的肝组织减卵率分别为34.2%、39.29%和43.66%,各实验组与对照组比较,经t检验差别有显著意义(P<0.05);各实验组间两两比较,经X^2检验差别无显著意义(P>0.05)。病理可见重组Sj-TPM组小鼠肝脏表面较光滑,质地较软,色泽略带暗红色,隐约可见少量灰白色小点,虫卵结节较少;生理盐水组小鼠肝组织中可见大量的虫卵肉芽肿形成,肉芽肿体积较大,周围有大量炎细胞浸润,较多的肝细胞变性坏死。结论日本血吸虫磷酸丙糖转移酶可能是一种潜在的血吸虫病疫苗候选分子。Objective To observe the immuno-protection in mice attacked with cercaria for a new vaccine candidate of Schistosoma japonicum,the recombinant protein triosephosphate transferase(SjTPM). Methods The BALB/c mice were immunized with the recombinant SjTPM in the dose of 50μg/ 100μl, 100μg/100μl and 200μg/100μl,the saline solution was used for control group,then attacked with cercaria. The percentage of worm reduction and egg reduction was calculated to evaluate the immuno-protection effect. HE staining method was used to detect the hepatic changes and granulomatous changes in the liver tissue. Results The percentage of worm reduction was 27.1%,32.1%and 35.2%,respectively,in the experimental groups.The number of eggs in liver tissue was reduced by 34.2%,39.29%,43.66%,respectively.There were remarkable difference among the experimental groups and control group by t -test.There was no remarkable difference between the experimental groups byχ~2-test.Gross observation showed that the livers in control groups were dark brown and hard,with miliary nodules of worm eggs diffused in the livers.The recombinant SjTPM group has some extent of amelioration,the livers were bright red,relative soft and smooth,with less nodules of worm eggs in livers.Hepatic histological section demonstrated that there' re generous large granulomas containing worm eggs in the livers of control group,with extensive infiltration of inflammatory cells and degeneration and necrosis of hepatic cells.The recombinant SjTPM vaccine group reversed the hepatic injury better than the control group.The structure of hepatic lobule was integrated on the whole,and the inflammatory reaction around the worm egg granulomas was light.Conclusion The molecular vaccine of rSjTPM had some immuno-protection effect.It suggested that rSjTPM was a potential candidate of S.japonicum.
