摘  要:目的:筛选梭果黄芪总黄酮的最佳纯化工艺。方法:采用静态与动态-解吸附方法对梭果黄芪总黄酮进行纯化,以吸附率与洗脱率等指标优化工艺,利用紫外-可见分光光度仪测定梭果黄芪总黄酮的含量。结果:X-5型大孔树脂纯化效果最好,其最佳工艺为上样液总黄酮浓度为4.05mg.mL-1,上样量为30mL,上样流速为1.5mL.min-1,分别以蒸馏水3BV、50%乙醇6BV按1.5mL.min-1的流速洗脱,弃去蒸馏水洗脱部分,收集50%乙醇洗脱部位。结论:该工艺操作简单,稳定可行,重复性好,适合梭果黄芪总黄酮纯化。Objective:To establish a technical process for purification of total flavonoids from Astragalus ernestii.Method:The optimal techniques of purification of flavonoid were obtained by static adsorption and elution tests,and the techniques were evaluated by measuring the concentration of total flavonoids from Astragalus ernestii with ultraviolet spectrophotometer.Result:The best effect of separation was found in the experiment with X-5 macroporous resin with the sampling rate 1.5 mL﹒min-1,concentration 4.05 mg﹒mL-1,sampling size 30 mL and 3BV respectively with distilled water,6BV 50% ethanol according to 1.5 mL﹒min-1 flow rate elution.The distilled water elution parts were adandoned,and collecting 50% ethanol elution parts were collected.Conclusion:X-5 resin can be applied to purify total flavonoids from Astragalus ernestii.
