摘  要:目的:优选瓦布贝母地上茎叶提取物的最佳纯化工艺。方法:以解吸液中的总生物碱含量为指标,考察大孔吸附树脂对瓦布贝母地上茎叶提取物的最佳吸附及洗脱条件。结果:以X-5大孔吸附树脂为最佳纯化树脂,上样液浓度为5.221mg/g、上样pH值为8.5,洗脱液为pH=3、浓度为75%的乙醇50BV冲洗大孔吸附树脂,提取物中总生物碱含量达到最高为18%。结论:该方法能明显提高瓦布贝母地上茎叶部分提取叶中的总生物碱含量,实验优选的工艺能够满足瓦布贝母地上茎叶提取物的纯化要求。Objective: To select the best purification process for the extract of Wabuensis Beimu.Method: The total alkaloid content of desorption liquid from macroporous adsorption resin were taken as indexes to investigate the optimal adsorption and elution conditions of extract of epigean stem and leaves from Wabuensis Beimu by macroporous adsorption resin.Result: With X-5 macroporous adsorption resin as the best purification resin,5.221 mg.g-1as the sample liquid concentration,8.5 as the sample pH value,3 as the eluent pH,75% as the ethanol concentration and 50 BV as the volume of eluent,the maximum total alkaloid content in extract was 18%.Conclusion: This method can obviously improve the total alkaloid content in extract of epigean stem and leaves from Wabuensis Beimu,and the optimized method is feasible to enrich and purify the extract of epigean stem and leaves from Wabuensis Beimu.
