穿蛭膏对实验性兔膝关节骨质增生细胞因子的影响

摘  要:目的探讨穿蛭膏对兔膝骨关节骨质增生模型动物关节液、血液中白细胞介素-1β(IL-1β)、肿瘤坏死因子-α(TNF-α)、一氧化氮(NO)的影响。方法采用Hulth模型建立兔膝骨关节骨质增生模型;将兔随机分为正常对照组、模型对照组、骨质增生一贴灵组、穿蛭膏组,正常和模型对照组不用药物,其余各组以相应的药物贴敷,末次用药24 h后,空气栓塞处死动物,观察关节软骨的变化并行Mankin's评分,用放射免疫法对各组关节腔液及外周血中IL-1β、TNF-α、NO进行检测。结果 Mankin's评分穿蛭膏组与其他各组有显著性差异(P<0.01,P<0.05),表明穿蛭膏有明显减轻关节软骨损伤作用;穿蛭膏能显著降低实验性兔膝骨关节骨质增生模型中血清和关节冲洗液IL-1β、TNF-α、NO的含量(P<0.05,P<0.01)。结论穿蛭膏可通过抑制炎性细胞因子(IL-1β、TNF-α)的水平、降低NO的含量等作用,保护关节软骨而对骨质增生起防治作用。Objective To explore the effect of Chuanzhi Gao on joint fluid,interleukin-1 beta(IL-1 beta),tumor necrosis factor-alpha(TNF alpha),nitric oxide(NO) in the rabbit model with knee joint hyperplasia.Methods The bone hyperplasia model was established by using Hulth in rabbit knee joint.The rabbits randomly divided into normal control group,model control group,hyperosteogeny yitieling group and Chuanzhi Gao group.Except the normal and model group,the other groups were administered Chuanzhi Gao or yitieling strapping on knee joint.The rabbits were sacrificed at the end of experiment.Articular cartilage changes were observed by Mankin's score,and IL-1 beta,TNF alpha,NO in articular cavity liquid and peripheral blood of were detected with radiation immune method.Results Mankin's score revealed that there were significant difference between Chuanzhi Gao group and other groups(P<0.01,P<0.05).Chuanzhi Gao could significantly decrease the IL-1beta,TNF alpha and NO content in serum and joint blunt washing liquid of knee joint hyperosteogeny model(P<0.05,P<0.01).Conclusion Chuanzhi Gao has prevention and cure function on articular cartilage and bone hyperplasia through inhibiting the inflammatory cytokines(IL-1 beta,TNF alpha) level and reducing NO content.
