摘 要:目的对八角莲化合物进行分离和鉴别,测定八角莲成分的抗癌活性。方法采用渗漉、萃取、层析、色谱等方法提取、分离、鉴别化合物,采用MTT法测定鬼臼毒素的抗癌活性。结果从中分离得到3个化合物。并运用质谱、核磁共振色谱等手段对化合物结构进行解析,最终确定化合物为4',5'-二去甲基鬼臼毒素、八角莲醇和鬼臼毒素。MTT法检测结果显示,不同浓度的鬼臼毒素处理SGC7901细胞24,48和72 h后,鬼臼毒素呈时间与剂量依赖性抑制SGC 7901胃癌细胞生长。结论八角莲含有4',5'-二去甲基鬼臼毒素、八角莲醇和鬼臼毒素等化合物,其具有SGC7901细胞增殖抑制作用。Objective To isolate and identify the active component of dysosma versipellis and determine its anticancer activity.Methods The percolation,solvent extraction,chromatography and gas chromatography were used to extract and identify the compound and MTT were applied to determine the anticancer activity of podophyllotoxin.Results Three compounds are isolated.Mass spectroscopy and nuclear magnetic resonance chromatography were made to analysis the compound structure,which confirmed as 4',5'-didemethylpodophyllotoxin,dysosmarol and podophyllotoxin.The MTT shows that the proliferation of SGC7901 gastric cancer cells could be inhibited by the podophyllotoxin of different dose at 24,48 and 72 h in the time and dose-dependent manner.Conclusion Dysosma versipellis contains the compounds such as 4',5'-didemethylpodophyllotoxin,dysosmarol and podophyllotoxin,it own inhibitory effect on proliferation of SGC7901.
