摘 要:目的:建立一种灵敏度高的高效液相色谱法,测定大鼠血浆中乙酰胆碱酯酶抑制剂N-甲基巴婆碱的浓度.方法:流动相为甲醇∶水∶三乙胺(65∶35∶0.1);血浆样品用乙酸乙酯萃取;检测波长:270 nm;流速:1.0 mL/min.结果:线性范围为10-400 g/mL,线性关系良好,最低检测限为5 g/mL.N-甲基巴婆碱日内RSD≤3.7%,日间RSD≤3.1%,提取回收率为98.03%~105.42%(n=5).结论:本方法灵敏度高,精密度、重现性好,适用于N-甲基巴婆碱在生物样本中的检测和药理、药动学研究.Objective: To determinate the plasma concentration of N-methyl asimilobine,an acetyl cholinesterase inhibitor,by HPLC in rat. Methods: Plasma samples were cleaned up by extraction with acetic ether. The chromatogram was monitored by UV detection at a wavelength of 270 nm. The mobile phase consisted of Methanol-Water-TEA(65∶35∶0.1). The flow rate was l.0 mL/min. Results: The linear range was 10-400 g/mL,the lower limit of detection was 5 g/mL. The intra-and inter-run precision were lower than 3.7% and 3.1%(n=5),respectively.The extraction recovery was 98.03%~105.42%(n=5).Conclusion: The method is shown to be accurate, repetitive, simple to operate, and suitable for monitoring biological sample,clinical pharmacology and pharmacokinetic study.
