HPLC法测定济川养生驻颜不老方药酒中芦荟大黄素和大黄素的含量

摘　　要：目的:测定济川养生驻颜不老方药酒中芦荟大黄素和大黄素的含量。方法:色谱柱为Ultimate XB C18(250mm×4.6 mm,5.0μm);流动相为乙腈-0.2%甲酸水溶液(75:25,V/V);体积流量1.0 mL﹒min-1;柱温30℃;检测波长254 nm。结果:芦荟大黄素和大黄素在10 min内达到分离,峰形尖锐对称,线性范围分别为1.01～16.08μg﹒mL-1和1.01～16.20μg﹒mL-1,平均加样回收率分别为99.49%和98.78%。济川养生驻颜不老方药酒中芦荟大黄素平均含量为15.41μg﹒mL-1,大黄素平均含量为1.87μg﹒mL-1;济川养生驻颜不老方药酒经水解处理后,芦荟大黄素的平均含量为15.46μg﹒mL-1,大黄素平均含量为2.78μg﹒mL-1。结论:济川养生驻颜不老方药酒中芦荟大黄素的含量高于大黄素;水解样品后大黄素的含量显著升高,表明大黄素部分以结合态形式存在于药酒中,实验结果为药酒的临床合理应用提供参考数据。Objective: To detect the content of aloe emodin and emodin in ‘Jichuan yang sheng zhu yan bu lao fang'medicinal liquor by HPLC. Method: The separation was performed on a Ultimate XB C18(250 mm×4.6 mm,5.0 μm)eluted with acetonitrile and 0.2% formic acid(75:25) at a flow-rate of 1.0 mL﹒min-1. The detection wavelength was set at 254 nm and the column temperature was set at 30℃. Result: Aloe emodin and emodin was separated in 10 min with good shape. The linear range were 1.01 μg﹒mL-1～16.08 μg﹒mL-1and 1.01 μg﹒mL-1～16.20 μg﹒mL-1respectively, and the average recovery were 99.49% and98.78%. The average content of aloe emodin and emodin in ‘Jichuan yang sheng zhu yan bu lao fang' medicinal liquor were 15.41μg﹒mL-1and 1.87 μg﹒mL-1. The average content of aloe emodin and emodin in hydrolysis sample were 15.46 μg﹒mL-1and 2.78 μg﹒mL-1. Conclusion: The Aloe emodin content in ‘Jichuan yang sheng zhu yan bu lao fang' medicinal liquor is higher than emodin.The emodin content in hydrolysis sample is higher than the sample without hydrolysis which indicates part of emodin exists as conjunctive situation. The experiment provides the reference for the clinical application.
