摘 要:目的 建立测定红景天苷含量的荧光分光光度法.方法 选择了荧光最佳的激发波长和发射波长,考察了放置时间、温度对测定结果的影响,给出最佳分析参数.结果 红景天苷的荧光最大激发波长λex=273 nm,最大发射波长λem=301 nm.红景天苷在0.01~0.10 mg/L范围内呈良好的线性关系(r=0.999 0).检出限DL=0.005 mg/L,0.05 mg/L的红景天苷相对标准偏差RSD=0.32%(n=5).且利用荧光分光光度法测定了西藏大花红景天提取物中红景天苷的含量.结论 荧光分光光度法灵敏度、快速,检出限低,可用于药材中红景天苷的含量测试.Objective To establish the spectrophotofluorimetry of determination the content of salidroside. Methods The optimum fluorescence excitation wavelength and emission wavelength was investigated. The effects of laying time and temperature were investigated and the optimum analytical parameters were obtained. Results The maximum ex- citation wavelength of salidroside was λ ex = 273 nm and the maximum emission wavelength was λ em = 301 nm. The lin- ear range of salidroside was 0. 01 ~ 0. 10 mg / L with the linearly dependent coefficient of 0. 999 0. The Detection Limit( DL) was 0. 005 mg/L and the Relative Standard Deviation( RSD) was 0. 32% for 5 independent determinations of 0. 05 mg / L salidroside solution. The content of salidroside in Rhodiola crenulata( from Xizang,China) was extracted and determined using the founded method. Conclusion The spectrophotofluorimetry was sensitive and rapid with a low detection limit. The method can be used to determine the content of salidroside in crude drugs.
