摘  要:基于H2O2 +4 - 氨基胺替吡啉+ 苯酚———醌式染料(λmax507nm) + H2O 反应,研究了不同过氧化物酶测定H2O2 的方法- 在pH7-02 Tris- HCl 缓冲溶液中,系统研究了辣根过氧化物酶(HRP) 及模拟酶的催化活性,其中β- 环糊精- 氯化血红素催化H2O2 氧化苯酚与4 - 氨基胺替吡啉生成醌式染料反应的催化活性最高,研究了β- 环糊精- 氯化血红素作为模拟酶的显色反应条件和在显色反应中β- 环糊精- 氯化血红素和底物的最佳浓度0~6-8×10- 5mol·L- 1 过氧化氢浓度范围测定具有好的重线性和精确度,表观摩尔吸光系数为1-10 ×104L·mol- 1·cm - 1-Various mimetic enzymes of peroxidase have been used a catalyst insted of peroxidase in determination of hydrogen peroxide based on reaction: H 2O 2+4-aminoantipyrine+phenol→quinon dye (λ max 507nm)+H 2O In the pH7 02 Tris-HCl buffer solution, the difference of relative catalytic activity was systematic studied between natural enzyme (HRP) and mimetic enzymes β-CD-hemin was the best one of the substitutions for peroxidase in determination of hydrogen peroxide by formation of dye from phenol and 4-aminoantipyrine The optimal concentration of mimetic enzyme and substrate were studied in the β-CD-hemin-Phenol-H 2O 2 system Meanwhile the optimal condition of color reaction was investigated 0~6 8×10 -5 mol·L -1 concentration rage of hydrogen peroxide was determinated with good accuracy and reproducibility The apparent molar absorptivity for hydrogen peroxide was 1 10×10 4L·mol -1 ·cm -1
