摘 要:目的 通过观察复合疲劳大鼠的空间学习、记忆能力及其海马区磷酸化环磷腺苷相应元件结合蛋白(phosphorylated cyclic adenosine monophosphate responsive element binding protein,p-CREB)的表达水平,探讨复合疲劳对大鼠学习记忆能力影响的机制. 方法 按随机数字表法,将24只成年雄性Sprague Dawley(SD)大鼠分为对照组、食物限制组和复合疲劳组3组,每组8只.运用Morris水迷宫实验方法测定各组大鼠的平均逃避潜伏期及平台所在象限的游泳时间百分比.运用蛋白质印迹(western blot)方法检测大鼠海马区p-CREB的表达.采用SPSS 17.0统计软件对3组大鼠的负重游泳时长、平均逃避潜伏期及平台所在象限的游泳时间百分比进行单因素方差分析. 结果 5d负重游泳实验结果显示,3组大鼠间游泳时长差异有统计学意义(F=40.28~66.04,P<0.01);与对照组和食物限制组相比,复合疲劳组大鼠的负重游泳时长明显缩短(P<0.01).在Morris水迷宫定位航行实验中,3组大鼠之间第2、3、4天的逃避潜伏期差异有统计学意义(F=4.17~7.11,P<0.05或0.01);与对照组和食物限制组相比,复合疲劳组大鼠的逃避潜伏期明显延长(P<0.05或0.01).在空间探索试验中,3组大鼠在平台所在象限的时间百分比差异有统计学意义(F=3.62,P<0.05);与对照组和食物限制组相比,复合疲劳组大鼠在平台所在象限的时间百分比明显降低(P<0.05).3组大鼠海马区p-CREB的相对含量差异有统计学意义(F=730.07,P<0.01);复合疲劳组大鼠低于对照组和食物限制组(P<0.01). 结论 复合疲劳对大鼠的空间学习、记忆能力具有损害作用,其机制可能与复合疲劳组大鼠海马区p-CREB的表达水平降低有关.Objective To explore the mechanism of cognitive change after complex fatigue by observing the effects of complex fatigue on learning and memorial abilities and on phosphorylated cyclic adenosine monophosphate responsive element binding protein (p-CREB) expression in hippocampus of rats.Methods Twenty-four adult male Sprague Dawlcy (SD) rats were used as experimental subjects.According to random number table,they were divided randomly into control group (n=8),food-restricted group (n=8) and complex fatigue group (n=8).All rats were subjected for Morris water maze (MWM) to test the mean escape latency and the percentage of time spent in the target quadrant.p-CREB level in hippocampus was tested with western blot.The software package SPSS 17.0 was applied for one-way ANOVA to analyze the weight-loaded swimming time and mean escape latency of three groups.Results The 5-d weight-loaded swimming test results showed that the swimming time of the complex fatigue group was significantly shorter than that of the control and food-restricted group (F=40.28-66.04,P<0.01); The mean escape latency in complex fatigue group from the second to fourth day of MWM was significantly longer than that in control and foodrestricted group (F=4.17-7.11,P<0.05 or 0.01); And in the spatial probe test,the rats in the complex fatigue group spent less platform quadrant time than that in control and food-restricted groups (F=3.62,P<0.05).Results of western blot for p-CREB revealed that the p-CREB level in hippocampus in complex fatigue group was significantly lower than that in control and food-restricted groups (F=730.07,P<0.01).Conclusions The impairment of spatial learning and memorial abilities in complex fatigue rats may relate with the reduction of p-CREB in hippocampus.
