摘  要:采用6对圆口铜鱼(Coreius guichenoti)微卫星引物对长江重庆、沙市、湖口和常熟4个江段的铜鱼(Coreius heterodon(Bleeker))的遗传多样性进行分析。PCR分析结果表明:6对引物均能成功扩增出DNA片段并具有一定的多态性,等位基因数在2~7个。每个群体的平均等位基因数为3.0~3.7,平均观测杂合度(Ho)在0.4844~0.6301,平均期望杂合度(He)在0.4695~0.5622,多态信息含量(PIC)范围在0.1638~0.7392。有部分微卫星座位在一些群体中偏离哈代-温伯格平衡。遗传距离分析表明沙市群体与常熟群体遗传距离最远,而重庆群体与湖口群体遗传距离最近。分子变异分析(AMOVA)表明,来自群体间变异占总遗传变异的0.59%,固定系数(FST)为0.0059,这表明长江水系铜鱼群体分化极小,可看作为一个群体。Six pairs of primers for microsatellite DNA of largemouth bronze gudgeon(Coreius guichenoti) were used to analyze the genetic diversity and characteristics of four populations of bronze gudgeon(C.heterodon)from the four sites along the Yangtze River,i.e.,Hukou(Jiangxi Province),Changshu(Jiangsu Province),Shashi(Hubei Province)and Wanzhou(Chongqing city).The results showed that the six pairs of primers were amplified successfully and had exhibited genetic polymorphism in bronze gudgeon(C.heterodon).The number of alleles in these loci ranged from 2to 7.The number of observed alleles of apopulation ranged from 3.00to 3.67on average,and mean observed heterozygosity(Ho)was 0.48to 0.63,while the mean expected heterozygosity(He)was 0.47to 0.56among the populations.The polymorphic information content(PIC)ranged from 0.163 8to 0.739 2.Several loci were found to be deviated from HardyWeinberg Equilibrium(HWE)in some populations.Genetic distance analysis showed that the distance between Shashi(SBG)and Changshu(CBG)populations was the farthest,while the distance between Chongqin(CQBG)and Hukou(HBG)populations was the nearest.The analysis of molecular variance(AMOVA)indicated that the Fixation Index(FST)was 0.005 9,meaning that 0.59%genetic variance occurred among the four populations.This showed that no genetic divergence had occurred at the level of nuclear DNA for bronze gudgeon(C.heterodon)in the Yangtze River.
