石斛合剂对STZ损伤胰岛细胞的保护作用

摘　　要：目的观察石斛合剂对胰岛细胞损伤的影响。方法分别用0%、5%、10%、20%的石斛合剂含药血清和13.3 nmol/L的IGF-1孵育原代培养7 d胰岛细胞24 h后,加入1.0 mmol/L的STZ作用24 h,采用MTT法观察胰岛细胞活性;AO-EB染色法观察各组胰岛细胞的存活情况。Annexin V-PI法检测各组胰岛细胞的凋亡、坏死情况。结果 10%、20%石斛合剂含药血清对胰岛细胞的保护作用显著,细胞活性和存活细胞数均较模型对照组明显增加(P<0.05),凋亡、坏死率较模型组明显下降(P<0.05),其保护作用与IGF-1阳性对照组相似。结论石斛合剂含药血清能保护胰岛细胞,减轻STZ对其所致的损伤。Objective To investigate the effect of Dendrobium compound(DC) on islet cells injuried by streptozotocin. Methods Primarily cultured cells were treated with 0,5%,10%,20% DC drug-containing serum and 13.3 nmol / L IGF-1 respectively for 24 hours,then the cells were treated with 1.0 mmol / L STZ for 24 hours. The activity of islet cells was observed by MTT assay,the survival condition of islet cells in each group was observed by AO-EB dyeing,and the apoptosis and necrosis of cells were detected by Annexin VPI methods. Results 10%,20% DC drug-containing serum had significantly protective effect on islet cells,the activity and survival rates of cells in DC group were increased significantly compared with model group(P&lt;0.05),and the rates of apoptosis and necrosis were decreased(P&lt;0.05). The DC group and IGF-1 group showed similar protective effects. Conclusion DC drug-containing serum can protect islet cells,and reduce the cells injury induced by streptozotocin.
